Short answer
Coen is human by day and a vampire at night. Human Coen uses a sword and rune magic; vampire Coen gains claws, supernatural powers and different movement options. Some NPCs and quests only appear at a particular time, and changing the time of day spends story time.

Day and night change more than combat
The time of day can change who is present, which quests appear and how Coen reaches an area. A missing objective may be waiting for the other half of the cycle rather than another quest trigger.
Do not switch only to test a form
The transition spends story time. Finish the free exploration available in the current form, then change when you have a clear destination or objective for the other one.

Return at the other time of day. The developers say some NPCs and quests are time-specific.
A route looks unreachable
Check whether a vampire movement ability opens it after dark.
